OECD no. 439: In vitro skin irritation - Test on reconstituted human epidermis

At SMC Laboratories, we are convinced of the quality of our products. However, we feel it's essential to carry out a few tests to make sure they're perfectly safe. So, in all transparency, here is the essential information.

 

WHY an in vitro test? 

Instead of testing on animals (or humans), in vitro laboratory testing, while more costly, is an excellent ethical alternative. An important value and proof of the Swiss Medical Cannabis teams' commitment to sustainable and ethically irreproachable development. Among the latest technological advances, in vitro tests are sophisticated models available to the parapharmaceutical industry. They are used to test products and their ingredients for safety (safety in use) and regulatory toxicity, as well as to test their efficacy and justify their promise of improved well-being. We have therefore opted for a protocol designed to measure the non-irritant potential of our product by means of an "in vitro" test on reconstructed human epidermis, in accordance with OECD guideline reference 439.

 

a precise methodology

The test guideline describes a procedure used to identify the hazards of irritant chemicals. As we explained, it is based on reconstructed human epidermis (RHE) or "laboratory reconstituted skin". Its design closely mimics the biochemical and physiological properties of the upper parts of human skin.

Cell viability is measured by the enzymatic conversion of the vital dye MTT to a blue formazan salt, which is measured quantitatively after tissue extraction. Irritant test substances are identified by their ability to reduce cell viability below defined threshold levels (less than or equal to 50% for UN GHS category 2).

This test guideline also includes a set of performance standards for the evaluation of similar and modified RhE-based test methods.
